SOMATIC EMBRYOGENESIS FROM ZYGOTIC EMBRYOS AND MERISTEMS OF FREESIA HYBRIDA

Abstract:
The regeneration of fertile plants from an embryogenic cultures of freesia has been achieved. Serially propagated embryogenic cultures were initiated from meristems and mature embryos of many genotypes. The initial explants produced embryogenic callus and/or embryogenic structures which differentiated from the cotyledon, epicotyl and hypocotyl or zygotic embryos as well as on the corm apical meristems. Histological evidence showed that some of the embryos arose directly from the explant. Genotypic variation in the frequency of incubation of embryogenic tissue was noted. Embryogenic tissue was chronically maintained on liquid or agar solidified MS medium with 2 mg 1-1 both of Picloram and 6-benzylaminopurine. Upon transfer to appropriate media somatic embryos in the callus as well as those formed directly on explants developed to maturity and germinated to produce green plants. Acceleration of embryo maturation was processed by transferring the tissue mass to liquid MS medium with 0.02–0.2 mg 1-1 Picloram and subjected to gyratory shaking. Whole fertile plants were recovered. Cultures have been maintained for 18 months without loss of morphogenic competency.
