METHODS TO DETERMINE ABSCISIC ACID AND INDOLE-3-ACETIC ACID, AND DETERMINATIONS OF THESE HORMONES IN TULIP CV. APELDOORN BULBS AS RELATED TO THE COLD-TREATMENT

Abstract:
ELISA's with monoclonal antibodies against abscisic acid (ABA) or indole-3-acetic acid (IAA) were developed. To determine these growth substances in tulip cv. Apeldoorn bulbs, extensive extraction procedures were necessary.

For IAA, the ELISA was insensitive in comparison with HPLC combined with fluorescence detection, and this last method proved to be more useful.

This way, we could determine the ABA and IAA levels in sprouts of uncooled, partly cold-treated and completely cold-treated (12 weeks, 5°C) tulip bulbs cv. Apeldoorn. The ABA levels per g FW did not change during this period, neither at 5 nor at 17°C. The IAA levels looked more promising.
