FACTORS INFLUENCING EX VITRO ROOTING OF TISSUE CULTURED PAPAYA SHOOTS

Abstract:
Factors influencing ex vitro rooting of tissue cultured papaya shoots were determined. The medium composition markedly affected in vitro shoot growth and degree of vitrification, and subsequently ex vitro rooting of the microcuttings. The medium containing one and half strength MS inorganic salts, 3% sucrose, 1.2% agar and 0.5 mg/liter 2iP was recommended to produce microcuttings which have higher potential to root ex vitro. Treating the microcuttings with auxin was highly effective for vigorous rooting, particularly with IBA at 2500 mg/liter. Vermiculite was the best rooting substrate under ex vitro condition.
