NITRIC AND AMMONIACAL NUTRITION IN TISSUE CULTURE: BEHAVIOUR OF THREE MAHONIA CLONES

Abstract:
Some genotypes of Mahonia appeared difficult to propagate through tissue culture.

A best knowledge of physiological behaviour and real nutritional requirements was attempted in order to improve mass-micropropagation of interesting clones of Mahonia. Metabolic study of nitrogen assimilation is reported for three clones in vitro cultured : Mahonia japonica 'Wintersun', M. aquifolium 'Apollo' and M. aquifolium 'Undulata'. The results show that in vitro, enzyme activity of GDH, linked to direct ammonium incorporation is predominent as regards to GS and NR activities associated to nitrate assimilation. A competition between nitric and ammonium uptake appeared.

The resuls already enable us to define for 'Apollo' a new culture medium with accurate supply of both nitrate and ammonium ions.
