Abstract:
Isolated microspores of Brassica crops such as Chinese cabbage, broccoli and rapeseed produce many embryos in vitro.
Desiccation tolerance of the embryos was induced by exogenous application of abscisic acid.
Cotyledonary stage embryos treated with 10–100mM abscisic acid acquired the highest desiccation tolerance and showed a 40% to 50% plant conversion frequency.
Desiccation tolerance was not dependent on species or the genotypes tested.
On the other hand, almost all embryos lost their viability after desiccation, when they were not treated with abscisic acid.
The dry embryos kept their conversion ability after six months of storage with no control of temperature and humidity.
The dry embryos had ability of plant conversion in soil conditions.
The plantlets recovered from dry embryos were not different from those from non-desiccated ones.
