IN VITRO PROPAGATION OF POPULUS SPECIES: RESPONSE TO GROWTH REGULATORS AND MEDIA COMPOSITION.

Abstract:
Physiological studies on the effect of growth regulators and media composition were conducted on in vitro propagated Populus tremula and P. trichocarpa as a basis for establishing in vitro propagation procedures. Benzyladenine (BA) induced extensive bud break of axillary buds, however root initiation was completely inhibited even at low concentrations. The effect of different auxins on growth rates and rooting demonstrated that the effects of hormone treatment in vitro were carried over to the hardening stage. Successful hardening of the plantlets in mist chambers was directly correlated to the previous hormone used in vitro. Shoot regeneration from root cultures of aspen demonstrates the possibility of producing rooted plantlets in one step.

Leaf yellowing and meristem dieback of P. trichocarpa in culture was significantly reduced by growing this species on half strength Gresshoff and Doy medium (1974) supplemented with Ca-gluconic acid and 2-[N-morpholino] ethanesulfonic acid (MES).
