Abstract:
Ex vitro derived leaf explants of sixteen cultivars of Gerbera jamesonii were irradiated with 5 – 25 Gy doses of gamma rays.
Dose rate was 1.92 Gy min-1. For the production of adventitious shoots, leaf explants were cultured on MS medium with 3 mg L-1 BA, 0.5 mg L-1 IAA, 100 mg L-1 myo-inositol, 0.4 mg L-1 thiamine·HC1, 10 g L-1 sucrose and 8 g L-1 Bacto Bacto-agar.
Shoot formation (frequency and intensity) was dependent on dose of irradiation and cultivar.
Doses of 20 and 25 Gy drastically restricted the regeneration ability of leaf explants, but even 25 Gy dose did not reduce the production of adventitious shoots completely.
