Abstract:
A novel disposable box-shaped culture vessel, the 'Culture Pack', was developed by using fluorocarbon polymer films (Neoflon® films) which possess superior properties such as thermal stability, high light transmittance and gas-permeability (Tanaka et al., 1988a). This paper first covers the shoot development of Spathiphyllum on rockwool (sixteen joined-blocks, 4 by 4, of Grodan® Multiblock AO 18/30) with liquid medium in the 'Culture Pack'. The growth of the plantlets on the rockwool in the 'Culture Pack' were normal and more vigorous when compared to those on agar medium in conventional glass bottles.
Attempts were also made to improve the photoautotrophy of in vitro shoots during the rooting phase on the rockwool with sugar-free medium in the 'Culture Pack' under CO2 enriched condition.
Plantlets grown on rockwool with sugar-free medium in the 'Culture Pack' under 3,000 ppm CO2 were significantly larger than those grown on sucrose containing agar medium in bottles under CO2 nonenriched condition.
CO2 enrichment for the 'Culture Pack'-rockwool system was found to contribute to an improved growth of the plants in acclimatization.
