IDENTIFICATION OF EIGHT JUGLANS SPP. AND THEIR INTERSPECIFIC PYBDRIDS BY ISOENZYMATIC ELECTROPHORESIS.

Authors:   E. Germain, I. Hanquier, R. Monet
Abstract:
Isoenzyme polymorphism was analysed in young leaf tissues from eight Juglans spp. (representing 28 clones) and their interspecific hybrids (representing 16 clones). Using vertical polyacrylamide gel electrophoresis nine enzymatic systems were identified which gave well resolved banding patterns (i.e., GDH, SKDH, TO, AAP, AMY, END, EST, LAP, GOT).

A narrow intraspecific variability was observed in the above enzymatic systems. This was probably due to the limited number of clones examined for each species. However isoenzymatic electrophoresis provided a useful diagnostic tool for identifying the different species and interspecific hybrids used. The use of isoenzymes as genetic markers is discussed and some genetic hypotheses are considered.

Abbreviations used: GDH, glutamate dehydrogenase: SKDH, shikimate dehydrogenase; TO, tetrazolium oxidase. AAP, alanine aminopeptidase: AMY, amylase: END, endopeptidase: EST, esterase: LAP, leucine aminopeptidase: GOT, glutamate oxaloacetate transaminase.
