ENZYMATIC CDNA AMPLIFICATION OF HOP STUNT VIROID VARIANTS FROM NATURALLY INFECTED FRUIT CROPS

Abstract:
Hop stunt viroid (HSV) cDNA reverse transcribed in vitro from total nucleic acid extracts of infected tissue was amplified using Taq I DNA polymerase and HSV group-specific DNA primers in a reverse transcription-polymerase chain reaction (RT-PCR). The RT-PCR assay is a rapid and specific method to determine HSV infection in fruit crops without viroid purification or the need for large amounts of infected tissue.
