SURFACE STERILIZATION FOR DIFFERENT PARTS OF THE SHOOT TIPS OF PRITCHARDIA ORNAMENTAL PALMS, PROPAGATED IN VITRO.

Abstract:
Pritchardia kaalae Rock seedlings of 2–3 years of age were taken from their growing medium (pots of peat and sand mixture), and in the laboratory roots and green leaves were removed and most of the cork. Shoot tips and leaf primordia surounded by white soft leaves were kept in an antioxidant solution in the refirgerator. Surface sterilization treatments applied on the explants were:
  1. Dipping in Ethyl alcohol 70% for 5 minutes (mins). (T1)
  2. Dipping in Chlorox 3% (commercial sodium hypochlorite, active ingredients 5.2%) for 20 mins. (plus Tween 20). (T2)
  3. Dipping in undiluted Chlorox for 20 mins. (plus Tween 20). (T3)
  4. Dipping in Ethyl alcohol 70% for 5 mins. then in Chlorox 3% (plus Tween 20) for 20 mins. (T4)
  5. dipping in Ethyl alcohol 70% for 5 mins. then in Mercuric chloride 0.3% for 5 mins. (T5) After each treatment explants were rinsed three times in autoclaved distilled water.
