PRACTICAL APPLICATION OF A NOVEL DISPOSABLE FILM CULTURE VESSEL IN MICROPROPAGATION

Abstract:
This paper first covers the shoot development of many horticultural plants on agar medium in the 'Culture Pack', which was developed as a novel disposable culture vessel made of fluorocarbon polymer film.
It was found to be suitable for shoot development of Cymbidium and Spathiphyllum (Tanaka et al., 1988a, b). The results show that the 'Culture Pack' is suitable and has no adverse effect on shoot development.
Attempts were also made to establish plantlets of several horticultural plants on artificial substrates, rockwool (16 joined blocks of Multiblock AO 18/30, Grodania A/S, Denmark) or a polyesterfiber block (Toyobo Co.
Ltd., Japan) with liquid medium in the 'Culture Pack'. Plantlets grown on rockwool in the 'Culture Pack' were normal, larger and more vigorous compared to those on agar medium or polyesterfiber block.
The optimal amount of liquid medium to be poured to rockwool in the 'Culture Pack' for shoot development, and the ease for transferring the plantlets grown in the 'Culture Pack'-Rockwool system to soil are also discussed.
