PLANT REGENERATION AND FIRST ATTEMPT OF IN VITRO GENETIC IMPROVEMENT OF OLIVE (CV MORAIOLO)

Abstract:
Plant regeneration from leaf petioles and leaf discs of olive (cv Moraiolo) and the possibility of genetic transformation, were investigated.

Shoot regeneration was obtained from leaf petioles in absence or in presence of a small amount of callus in auxin-free MS basal medium supplemented with Thidiazuron (TDZ) 5.0 μM (regeneration 20%) or 20.0 μM (regeneration 10%). Shoot regeneration from leaf petioles with formation of larger callus (10%), was obtained on the same MS basal medium supplemented with TDZ 10.0 μM in combination with NAA 2.5 μM. Leaf disc did not give regeneration.

Transformation of olive cultivars, by using A. tumefaciens strain LBA 4404 containing the vector Bin 19 in which an A. rhizogenes T-DNA Eco R1 fragment carrying the "root inducing" rol B gene had been cloned, was attempted by punctured with infected needle axillary buds or by co-culturing leaf petioles.

On selection medium with Kanamycin, shoots from axillary buds did not grow, whereas the petioles produced shoot like structures that survived for a long time but did not evolve into well growing shoots.
