GENETIC STABILITY AND EXPRESSION IN CULTURED CELLS OF NICOTIANA TABACUM L. ADAPTED TO OSMOTIC STRESS

Abstract:
The development of methodology for selecting in vitro cellular clones of Nicotiana tabacum L. tolerant to the osmotic stress is described. Pieces of callus of four genotypes of Nicotiana tabacum L. have been put to different levels of osmotic pressure, adding up to the medium Polyetyle ne glycol 6000 in an increasing bound (0%, 15%, 20%). The cellular clones, which regenerated plants, are resulted those from androgenetic line 467HI/87 at the concentration of 15% or 20% of PEG. The regenerated plants from selected clones from the 1st cycle have been cultivated in vitro. From these plants has been made a tissue culture. Cellular clones submitted to the 2nd cycle of selection at reduced osmotic potential by addition of PEG with an increasing concentration (0%, 15%, 20%). After the treatment, the cellular clones have been placed in culture in the medium without PEG. In this way we regenerated plants from the subclones isolated at selective increased pressure. The plants regenerated from the selected clones of the original line 467HI/87, confirmed the stability and expression of tolerance the osmotic stress, it depends on the level of selecting pressure.
