GRAFTING ADULT SCION ON THE GERMINATED SEEDS OF CHESTNUT EXHANCES EXPLANT SURVIVAL

Abstract:
Two selected types of chestnut Castanea sativa Mill., were grafted on germinating chestnut seeds. The grafting took place on the hypocotyle, by using the technique of kerfgrafting. This was practised repeatedly for three consecutive years, whereafter explants were taken from the sprouting shoots, except at the last grafting: in that case the explants were taken from the shoots of four week old grafts. The sterilized explants were put on a modified Woody Plant Medium with a double concentration of Ca and Mg salts, vitamines according to Skoog, BA 0,2 mg/1 and IAA 0,1 mg/1. The explants from the four weeks old grafts showed all features that are typical for adult chestnut forms, i. e. rapid greying of the medium and an insignificant number of surviving explants. More surviving explants could be observed when shoots were harvested from the one year old grafts while the two years old grafts demonstraded the highest number of survival.

Reversion to juvenilits is more pronounced in explants taken from one or two year old grafts than it is in those that are four weeks old. We conclude that phase reversion requires a longer period of time, at least one growth season.
