MULTIPLE SHOOTS AND CALLUS FORMATION FROM SEEDS OF MANGOSTEEN (GARCINIA MANGOSTANA L.) CULTURED IN VITRO

Abstract:
Mature seeds of mangosteen (Garcinia mangostana L.) were segmented and cultured on MS and modified MS (with half-strength macroelements) media supplemented with combinations of NAA at concentrations of 0 and 2.5 μM and BAP at concentrations of 0, 30, 40 and 50 μM. Seeds cut into six segments produced more shoots than those cut into three segments. Cultures on MS medium under 8 hour photoperiod produced more shoots than other cultures. Medium supplemented with 40 μM BAP without NAA appeared to be the best for producing multiple shoots. Shoots were rooted, and plantlets formed were transferred into vermiculite and sand mixture(1:1). For callus formation, MS medium was supplemented with 2,4-D at concentrations of 0,20 and 30 mgl-1. Callus was observed to have been formed mainly from segmented seeds in cultures supplemented with 30 mgl-1 2,4-D under 12 hour photoperiod.
