SOMATIC EMBRYOGENESIS AND PLANT REGENERATION IN MAIZE

Abstract:
Callus cultures were established from immature glumes of maize genotypes on MS media with 2 mg/1 2,4-D. Calli grown on MS 2 media underwent embryogenesis and formed type I embryogenic calli. Plant regeneration was achieved upon transfer to media with low 2,4-D or with no hormones. Sucrose (2–3%) and higher ammonium nitrate enhanced the frequency of E calli. Biochemical characterization of embryogenic calli revealed specific peroxidase, esterase and MDH isozyme profile.
