TISSUE CULTURE AND GENETIC TRANSFORMATION OF DANDELION

Abstract:
A rapid and high frequency regeneration system (above 95%) for dandelion (Taraxacum mongoliam Hand Mazz) was obtained when petiole explants were cultured on MS medium containing 0.2mg/l IAA with 1 mg/l TDZ or BAP. This system was used for transformation by Agrobacterium Tumefaciens strain EHA101 carrying the binary vector PROA93 which contains 35S–GUS and 35S–NPT–II genes. Transgenic plants with KmR resistance and positive GUS activity have been recovered.
