IN VITRO CONSERVATION OF HAPLOID AND DIPLOID GERMPLASM IN ALLIUM CEPA L.

Abstract:
A collection of in vitro clones derived from gynogenetic haploids, diploid regenerants, and their mother plants is being established. Long term maintenance of this material comprises a cycle consisting of a slow growth phase at 4 °C and a multiplication phase using cuttings of in vitro bulblets and cultivation on media with benzylaminopurine (BAP). Maintenance of the haploid constitution is repeatedly checked by stomata guard cell measurements.
