RESEARCHES ON OBTAINING DIPLOIDE PLANTS IN VITRO FROM THE PISTILS OF NICOTIANA RUSTICA L. |

| Abstract:
For these researches, we took the boutons of N.rustica L. ten day before flowering. We sterilized them in ethyl alcohol 70 % during 30 seconds and in sodium hypochlorite of 3 % during 35 minutes. We rinsed them with distiled water and sterilized 4 times during 5 minutes. Later, we took out only the pistils and put them in Murashige and Skoog medium which contain two sorts of composition of hormone: first solution contains 4 mg/1 of kinetine and 1 mg/1 IAA; the second contains 4 mg/1 of K and 1 mg/1 naphtylacetic acid. Finally, we obtainede diploide plants only from the stumps of IAA media. | |
