THE RESEARCHES OF OBTAINING FROM PISTACIA VERA L. APICAL BUD AND NODAL BUD BY MICPROPAGATION

Abstract:
At first, mature nuts have been selected. The nuts were surface sterilized in 70% ethanol for 30–40 s then 3 % NaOCl for 30 min. after three rinses in sterile distilled water. They were placed in the 10 % jeloz medium. Seedlings could be observed in a week or two from Pistacia vera nuts. Epicotyl zone were placed in MURASHIGE and SKOOG (1962) medium supplemented Kinetin (4 mg/l), NAA (1 mg/l). Shoot growth and proliferation could be observed in a month or two of the application of apical bud or nodal bud explants from Pistacia vera seedling. Shoots produced in vitro have been rooted on MS medium having absence hormone. Plantlets were produced and readily applied in peat-beased compost.
