MICROPROPAGATION OF HOLY (ILEX AQUIFOLIUM)

Abstract:
We have developed for the first time a procedure for holly micropropagation. Buds from adult holly trees were used as explants, surface sterilized, were inoculated on Murashige and Skoog (MS) culture medium. Several citokinins were used for auxillary bud induction on explants. BAP was found to the most effective with higher multiplication rates.

Axillary shoots were elongated. Best results in root production were achieved with pretreatment of 4h with high concentrations of NAA and further culture on 50% MS salts for six weeks. Healthy holly plantlets were recovered and transfered for ex vitro conditions in the green house.
