Abstract:
Shoot differentiation was obtained from embryonal explants of Pinus halepensis Mill. when cultured on cytokinin-containing media.
The concentration and duration of application of N6- benzyladenine (BA), the use of other cytokinins alone or in combination with BA, the basal media, and the age of explants all significantly affected the bud-forming capacity.
Shoot elongation was achieved on hormone-free medium and the elongated shoots were rooted by pulsing them with 1 mM indolebutyric acid (IBA) for 6 h.
At least 90% of the shoots formed normal roots and the rooted shoots were readily hardened and transplanted to the greenhouse with 100% success.
