ISHS Acta Horticulturae 289: International Symposium on Plant Biotechnology and its Contribution to Plant Development, Multiplication and Improvement

IN VITRO REGENERATION OF SHOOT BUDS AND PLANTLET FORMATION FROM MATURE EMBRYOS OF PINUS HALEPENSIS MILL.

Authors:   M. Lambardi, K.K. Sharma, T.A. Thorpe
Abstract:
Shoot differentiation was obtained from embryonal explants of Pinus halepensis Mill. when cultured on cytokinin-containing media. The concentration and duration of application of N6- benzyladenine (BA), the use of other cytokinins alone or in combination with BA, the basal media, and the age of explants all significantly affected the bud-forming capacity. Shoot elongation was achieved on hormone-free medium and the elongated shoots were rooted by pulsing them with 1 mM indolebutyric acid (IBA) for 6 h. At least 90% of the shoots formed normal roots and the rooted shoots were readily hardened and transplanted to the greenhouse with 100% success.
