MINERAL MEDIA FOR IN VITRO PROPAGATION OF JUVENILE ‘PICUAL’ MICROCUTTINGS

Abstract:
This work reports about a section of a research project aimed at setting up a micropropagation method for Spanish olive tree cultivars. Not one of the mineral media tried included growth regulators, vitamins or others addenda. Two series of trials were carried out, the first using eight mineral media as follows: MS, 50% MS, 25% MS, 0% MS (in Macroelements), N30Ca, N30K, N45K and WPM. The second checked out the best three (N30CA, N30K and 50% MS) versus OM (Rugini, 1984). One bud, half-leaf microcuttings taken from juvenile seedlings obtained by germinating endocarp-free ‘Picual’ seeds from a single tree, 30 test tubes per medium, were used. New rootless explants, obtained from each tube were transfered to fresh mineral media at 8, 14, 20 and 26 weeks after the initial establishment of the first microcuttings. Results show that ‘Picual’ is better adapted to the specific olive medium developed by Rugini (1984).
