INDUCTION AND USE OF VARIATION IN POTATO WITH DIFFERENT IN VITRO SYSTEMS

Abstract:
Irradiation of in vitro grown plantlets and subsequent micropropagation induced 9% of morphological variants, 76% of which were uniform. A very low rate of gross aberrants was found. In contrast, regeneration from mesophyll protoplasts gave rise to a much higher amount of variants (34%), many of them gross aberrants (43%) or chimeras (16%). Regeneration from tissue explants produced 24% of variants, with a small portion of gross aberrants (11%) or chimeras (19%). The irradiation of micropropagated plants appears to be an easy and efficient method for induction of mutation.

We challenged 1200 mutagenized potato shoots with culture filtrate of Phytophthora infestans and obtained 5.6% insensitive variants.
