IMPROVING YIELD OF ADVENTITIOUS SHOOTS IN APPLE

Abstract:
In vitro adventitious regeneration in apple cultivars 'Granny' Smith', 'Mark', 'Novole', 'Lancep' and 'Cepiland' was obtained. I report a significant increase in the number of regenerated shoots from 'Gala' and 'Golden Delicious'. Plants were regenerated both from callus or directly from leaves from micropropagated plants. For somaclonal variation studies, callogenesis was induced from leaves and regeneration was attempted from primary and secondary calli. Twenty one hormonal combinations were tried. For 'direct' adventitious regeneration, leaves were collected and cultured on different media and in light or in darkness. Fourty two hormonal combinations were tried. Organogenesis did not occur without cytokinins and was enhanced in darkness. One treatment gave, on 'Gala', 100 % regenerating leaves with an average of 14.2 regenerated shoots per leaf. Two thousand plants from 'Gala' were successfully acclimatized and 1000 of them are now entering field trials to evaluate their phenotypic uniformity.
