Abstract:
Protoplasts were isolated from leaves of in vitro rooted shoots.
Cultures gave high yields of protoplasts when incubated in the enzyme solution at 28°C for 18 h in darkness.
Initial cell division occurred after 1 week in protoplasts plated at 5 x 104/ml in gelled media, particularly in zeatin-enriched media, and cultured in darkness at 25 ± 1°C. After 20 days only occasional divisions were found in liquid media.
In contrast, growing cell-colonies were visible in solid media.
