CELL ENGINEERING BY PROTOPLAST FUSION: IMPLICATIONS FOR PLANT IMPROVEMENT

Abstract:
The transfer of cytoplasmic genomes from Lycopersicon and Solanum species to L. esculentum was attempted by means of the "donor-recipient" method. A cytoplasmic-albino Large Red Cherry mutant (ALRC) was utilized as the recipient genotype. Protoplasts from cytoplasm-donor genotypes were irradiated with X-rays, in order to induce chromosome elimination and transfer of cytoplasmic genophores upon fusion.

Putative cybrid colonies were isolated and subsequently regenerated. Characterization of ALRC x L. peruvianum cybrids revealed isozyme patterns identical to L. esculentum.. Cybrid plants exhibit a recombinant mt-DNA pattern, containing parental as well as novel fragments.
