Abstract:
Undeveloped ovules were extracted from ripe fruits of sweet orange |Citrus sinensis (L).Osb.| cvs 'Tarocco', 'Moro', 'Belladonna', 'Navelina' and 'Navelate'. The media used were Murashige and Tucher (MT) alone or supplemented with 6-benzyladenine (BA) and/or kinetin at 2.5, 5 or 10 mg/l.
Ovules of 'Tarocco', 'Moro' and 'Belladonna' oranges cultured on MT supplemented with 10 mg/l of BA alone or 5–10 mg/l of BA and kinetin together showed the highest percentage of embryogenic callus.
The friable embryogenic calli were subcultured for two years on MT medium alone or with 10 mg/l BA or 10 mg/l kinetin which maintained friability and embryogenic potential.
Calli transferred on MT medium supplemented with 50 g/l of lactose or 20 ml/l of glycerol gave rise to several somatic embryos and plantlets.
