ADVENTIVE REGENERATION IN CELL COLONIES OF CARNATION

Abstract:
A method to obtain shoot regenerations from callus and cell culture of carnation (Dianthus caryophyllus) is described. Fragments of in vitro multiplied plantlets of carnation were cultured on a solid medium based on Murashige and Skoog salts plus 1 ppm each of BA and NAA. The frable callus obtained was cultured in liquid medium with 7 ppm BA and 2 ppm NAA to induce cell proliferation.

The highest viability was obtained in the first and second day of culture but good viability was maintained for 10 days by reneuwing the medium. Cells and calli plated in Petri dishes with certain hormonal combinations showed good regeneration, but calli produced by using 2,4-D did not show regeneration. Resulting shoots were micropropagated and acclimatized to the greenhouse.
