Abstract:
Density separation of celery somatic embryos (Apium graveolens cv.
Utah Improved #15) cultured in liquid medium was achieved with layered sucrose gradients (9 to 40%). Embryos located in the interface between 9 and 12% sucrose had the highest fatty acid content and the highest survival after 24 hr desiccation at 90% relative humidity.
Desiccation tolerance was increased by supplementation of embryo production medium with 10-6M abscisic acid (ABA) or 1.0 mM proline.
Highest survival was achieved by a combination of ABA plus proline.
ABA increased the accumulation and altered the distribution of fatty acids in somatic embryos.
