Abstract:
We have evaluated the use of cotyledons as starting material to induce somatic embryogenesis in two cultivars of Castanea sativa and of two hybrid cultivars of C. crenata x C. sativa.
Somatic embryos were obtained, in darkness, only from immature 'G9' cotyledons cultured on half-strength Murashige-Skoog (½ MS) medium (1962) supplemented either with NAA alone or 2,4-D plus BA but not with B5 medium (Gamborg et al., 1968). Somatic embryos developed slightly when transferred to MS plus BA, GA3 and IBA, but well-developed seedlings were not observed.
