Abstract:
Conditions for culturing triploid calli from endosperm tissue of tomato (Lycopersicon esculentum Mill) are described.
Endosperm excised from nearly mature seeds is easily separated from diploid tissue and shows the highest rate of callus production.
When mature seeds are used, soaking in water for three days prior to endosperm excision enhances rates of callus formation.
Initiation of callus often occurs near the micropyle.
