Abstract:
A method to induce somatic embryogenesis in Genista monosperma Lam. by callus culture is described.
The callus developed from immature zygotic embryos (0.3 cm. average length) collected 10 weeks after flower pollination, and cultured on a Murashige and Skoog medium, containing 5 mg/l of 2,4-D (2,4-dichlorophenoxyacetic acid) and 0.5 mg/l of BA (6-benzyladenine). Embryogenesis from undifferentiated callus started on a medium composed of Murashige and Skoog (MS) macro-elements and micro-elements, glycine 2 mg/l, calcium pantothenate 1 mg/l, thiamine-HCl 1 mg/l, myo-inositol 200 mg/l, zeatin ( 6–4- hydroxy-3-methylbut-2-enylamino-purine) 5 mg/l, NAA (1-naphthaleneacetic acid) 0.1 mg/l.
Attempts to induce germination of somatic embryos were unsuccessful.
