ORGANOGENESIS IN CALLUS CULTURES OF PINEAPPLE (ANANAS COMOSUS (L.) MERR.)

Abstract:
Callus was induced from the crown apical region of 'Queen' pineapples on Murashige and Tucker medium with casein hydrolysate (400 mg/l), coconut water (15 %) and naphthaleneacetic acid (40 mg/l). Callus did not become organogenic unless it passed through a stage where the colour changed from yellow to green. By investigating the anatomical changes in the green callus it was possible to determine that the regeneration of plants was by indirect adventitious organogenesis, and not the result of somatic embryogenesis. Areas of meristematic activity were easily discernible, and developing shoot buds could be seen on the periphery of the callus as well as within the callus mass.
