Abstract:
A genomic library of E. amylovora wild-type isolate T was prepared in the cosmid pLAFR3 and mobilised into the capsulated mutant P66 which, unlike T, fails to cause fireblight symptoms in pear or the HR in beans or tobacco.
Three clones were identified which restored virulence and ability to cause the HR to mutant P66. Subcloning and transposon mutagenes is located the determinant restoring virulence to P66 on a 2.1kb BamH1/HindIII fragment which was common to each clone.
This fragment also restored ability to cause the HR. The three cosmid clones were also found to complement avirulent mutants of isolate T recovered following treatment with nitrosoguanidine.
The patterns of complementation observed indicated that a cluster of genes spanning c. 20kb controlled pathogenicity.
