DETECTION OF LATENT FUSARIUM OXYSPORUM IN GLADIOLUS CORMS

Abstract:
A laboratory test to detect (latent) infections of Fusarium oxysporum in gladiolus is being developed. Corms sealed in plastic bags together with a liquid selective medium were hammered to pieces and incubated at 30°C. After one week a 25-μl droplet was taken from each bag and put on an agar plate containing the same selective medium. Plates were incubated for 3 days at 30°C and then screened for the presence of colonies of Fusarium. Experiments involving addition of spores of F. oxysporum to the test system showed that very small numbers of propagules can be detected. When the results of the laboratory test were compared with the incidence of plants showing Fusarium symptoms, after corms from the same lots had been planted and grown under disease-favouring conditions, a good correlation was found. The possibilities to develop this laboratory test to a practically applicable one are discussed.
