INFLUENCE OF CULTURE ENVIRONMENT ON SHOOT GROWTH AND BULBING OF TULIP IN VITRO

Abstract:
Shoot regeneration from tulip floral stem explants was not influenced by the ethylene sources, 1-aminocyclopropane-1-carboxylic acid (ACC) and 2-chloro-ethylphosphonic acid (Ethrel), however bulbing of shoots was enhanced by ACC. Bulbing was also improved by incubating shoot cultures under lights with a low red/far-red ratio. Incubation at low temperature (4°C) and on media containing elevated levels of sucrose was found to enhance bulbing and was associated with higher levels of sucrose in the tissues. In contrast, good shoot regeneration appeared to be associated with elevated levels of glucose in the tissues.
