FURTHER CHARACTERIZATION OF ALCOHOL DEHYDROGENASE IN STRAWBERRY CALLUS

Abstract:
Strawberry (Fragaria x. ananassa cv. Allstar) callus derived from leaf explants and grown on Murashige and Skoog medium was used to characterize and study the activity of alcohol dehydrogenase (ADH). The enzyme from callus has been partially purified. This purified extract was used to determine kinetic constants for the substrates NAD and ethanol, and to obtain an estimation of the molecular weight. The enzymes banding patterns were visualized on polyacrylamide gels and have shown a greater intensity when ADH was extracted from callus subjected to low oxygen compared to control callus.

Individual cells, derived from callus that had been treated several months earlier with N2, were tested for tolerance to low O2 conditions. Cell cultures were treated with 2% or 6% O2 and viability was measured using various staining techniques. Data concerning this adaptive response will be presented.
