IDENTIFICATION OF RASPBERRY CULTIVARS BY STARCH GEL ELECTROPHORESIS AND ISOENZYME STAINING

Abstract:
Starch gel electrophoresis and isoenzyme staining were evaluated for the characterization of eighteen red and two purple raspberry (Rubus idaeus L. and R. x neglectus Peck) cultivars. Raspberry tissues were electrophoresed using five different gel/buffer systems and stained with up to fourteen separate enzymes. Five of these enzymes gave consistent staining and also showed polymorphism. Using these five enzymes fourteen cultivars were uniquely separated while the remaining six cultivars were assigned to three different groups. Freezing wicks imbibed with crushed leaf extracts at -20°C for 1 week prior to electrophoresis had virtually no effect on subsequent isoenzyme staining but longer storage reduced the visibility of bands in the gels.
