Abstract:
Shoot explants of Brassaia actinophylla Endl. cv.
Amate containing apical meristems were cultured on Murashige & Skoog medium (1962) supplemented with Thidiazuron and 6-benzyladenine at concentrations between 0.5–5.0 mg/l in combination with naphthaleneacetic acid at 0.10 or 0.25 mg/l.
At all tested concentrations, thidiazuron exhibited cytokinin activity, strongly inhibited root formation and stimulated shoot bud organization on explant tissue.
The lowest thidiazuron concentration tested (0.5 mg/l) was to high for effective shoot multiplication due to induced deformations. 6-benzyladenine was amenable cytokinin to adventitious shoot induction and the optimal concentration in multiplication medium was established as 3.5 mg/l with naphtaleneacetic acid at 0.10 mg/l.
Regenerants derived on medium containing optimal 6-benzyladenine dose were easily rooted in vitro and established in soil.
