Abstract:
Direct immunofluorescence microscopy is introduced as a fast and accurate method for diagnosing cymbidium mosaic (CyMV) and odontoglossum ringspot (ORSV) virus infections in fresh orchid tissues.
Non-specific immunofluorescent reactions were reduced considerably when tetramethyl rhodamine isothiocyanate- conjugated antiserum incubated with the healthy plant extracts before tissue sections stained.
A method of protein A-gold labelling is also described.
The application of the highly sensitive immunogold labelling provided a precise identification of ORSV- and CyMV related antigens.
Very specific gold labelling of the viral structures has been shown in the sections which have been treated with homologous antiserum, but did not in sections treated with either pre-immune serum or heterologous antiserum.
