Abstract:
Ornithogalum and Lachenalia species, which have been commercially grown as ornamentals in South Africa since the 1960's, are often infected with Ornithogalum mosaic virus (OMV). OMV is a filamentous virus and causes severe mosaic symptoms and flower deformation.
An enzyme-aided purification protocol was developed which eliminated a highly viscous mucilage from plant sap.
Biological, serological and physicochemical characterization of South African virus isolates from both genera confirmed that the virus is OMV and that OMV is a Potyvirus (Burger and Von Wechmar, unpublished results). Serological detection of OMV by DAS-ELISA and DOT-ELISA in post-dormant bulb tissue was attempted.
Results indicated that virus was concentrated in bulb growth points.
An assay utilizing small sections of leaf tissue from young plantlets, was devised.
This test entails a one-step extraction procedure and DAS-ELISA.
