DEVELOPMENT OF A PHOTOAUTOTROPHIC TISSUE CULTURE SYSTEM FOR SHOOTS AND/OR PLANTLETS AT ROOTING AND ACCLIMATIZATION STAGES

Abstract:
The prototype of a novel tissue culture system was constructed that could be used to culture shoots and/or plantlets photoautotrophically at rooting and acclimatization stages, and that could be used to provide a labor-saving. The photoautotrophic tissue culture system (PTCS) is composed of a growth chamber and the device which consists of a culture box assembly, a gas flow assembly, and a culture solution flow assembly. With the PTCS, shoots and/or plantlets are cultured under sugar-free, CO2 supplied, a high photosynthetic photon flux, a low relative humidity, and non-aseptic conditions.

A trial experiment showed that the averages of the dry weight and net photosynthetic rate of strawberry plantlets cultured with the PTCS were heavier and higher than those cultured with a usual tissue culture method for rooting stage, respectively. The results indicate that the PTCS is beneficial to photoautotrophic culture for strawberry shoots and/or plantlets.
