PHOTOSYNTHESIS OF REGENERATED PLANTLETS IN VITRO, AND THE STRESSES OF TRANSPLANTING

Abstract:
Following transplanting from in vitro culture to soil there are plant species whose in vitro-derived foliage cannot successfully adapt to photoautotrophy. The activity of the carbon-fixing enzyme RubPcase in the in vitro foliage of such species is significantly impaired by exogenous sucrose in the culture medium. There also appears to be no adaptive response once the sucrose is removed to sustain the plant following transfer to soil. There is evidence elsewhere that exogenous sucrose also affects RubPcase activity in photoautrophic suspension cell culture, and that the role of PEPcase as a carbon-fixing enzyme becomes more significant under these conditions. The photosynthetic responses of differentiated plantlets in vitro and following transplanting are discussed in this context.
