Abstract:
Tulip leaves of cv. 'Golden Age' were disrupted by blending in dry ice.
Nuclei were isolated after dehydration in cold acetone by centrifugation in carbon tetrachloride-cyclohexane mixtures of varying densities.
The purest nuclear fraction was obtained at a density between 1. 450 and 1. 470 at 0°C. DNA assays indicate about a five fold purification of nuclei Soluble proteins, histones, chromosome residual proteins, and nuclear residual proteins were prepared and analyzed for protein and DNA content.
