Abstract:
Juglans regia L. embryonic axes, the basal and apical sections derived and juvenile nodal segments were used as primary explants.
Tissues were cultured on a modified MS medium (1) in the presence of different BAP concentrations (0,0.5,1,2,4 mg/l). All material assayed responded to the presence of BAP 1 mg/l.
Elongation could be achieved by transfer to a medium wich reduced BAP and by a possible synergic effect between apical bud removal and BAP add.
Different auxins IBA, IAA, NAA in different concentrations were applied to facilitate plantlet regeneration.
While immersion of the basal end of embryonic axis in IBA 1 mg/l solution for 1 min was efficient; similar results were not reproductible for single shoots derived from proliferation clusters.
With this material the use of a solution with 1 mg/l of NAA for 5 min provided rhizogenic response on 42.8% of the shoots.
