IN VITRO CULTURE FOR CITRUS MICROPROPAGATION.

Abstract:
The "in vitro" propagation of the citrus rootstocks "Carrizo" and "Troyer" citranges(Citrus sinensis(L.) Osbeck x Poncirus trifoliata (L.) Raf.), a dwarf mutant of "Troyer" citrange of the citrumelo CPB 4475 rootstock(Citrus paradisi Macf. x P.trifoliata(L.) Raf.) and of the "Flying Dragon" trifoliata orange (P.trifoliata var.monstrosa(T.ITO) Swingle) are reported. Between the various media used, the highest rate of shoots multiplication was as follows:
  1. for the "Troyer" and "Carrizo" citranges and the citrumelo CPB 4475-MS basal medium supplemented with 6-BAP 1 mg/1, IBA 0.5 mg/l and adenine 40 mg/l.
  2. for the "Flying Dragon" and the "Dwarf mutant" - MS basal medium supplemented with 6-BAP 0.5 mg/l, IBA 0.25 mg/l and adenine 40 mg/l.

The best rooting was obtained, for all the four rootstocks and also for the "dwarf mutant" employed in the tests, utilizing the basal MS medium supplemented only with NAA 1 mg/l. The rooted young plants when cm 5 high were transplanted in small plastic containers filled with a steam sterilized mix of light vulcanic soil and peat moss. After transplanting, the young plantlets were raised in a hotbed frame. Utilizing the above mentioned technique for transfering the plantlets from "vitro" to the soil mixture only a limited number of them were lost(2%).
