Abstract:
In vitro rescue of immature, embryos is often the only way to achieve germination of early ripening peach varieties (cvs Armking and Maycrest). Two basal media were employed: Murashige e Skoog (MSe) and Stewart and Hsu (SHe) and according to their size the embryos were divided into 5 classes and cultured at different sucrose concentrations.
As the embryos reached a larger size they were transferred to a medium with a lower sucrose concentration.
For the two genotypes no appreciable difference was show in response to the different media.
The initial embryo size strongly affected its further growth and germination.
More than 50% of the embryos showed partial or disordered germination.
In order to save this material the entire cotyledons and the root primordia were dissected and the caulinair apex or meristem was cultured and treated as a normal peach shoot tip.
