AUTOMATION OF TISSUE CULTURE MANIPULATIONS IN THE FINAL STAGES

Abstract:
The major cost in plant propagation by tissue culture is manual labor. In order to save manual labor a production scheme is presented where the in vitro manipulations are limited to only one inoculation of plant material on a fresh solid multiplication medium.

A further step to reduce manual labor was the automation of the adding of the liquid medium. A transportable injection apparatus was developed which allows to inject the liquid medium directly through the lid of the plastic container.

With this production scheme Prunus avium plantlets were transfered to the greenhouse 10 weeks after inoculation on a multiplication medium, and liquid medium was injected twice to induce elongation and rooting. After two months of weaning survival rate was 95 %, and 8.7 plantlets with a mean lenght of 4 cm developed out of one initial explant.
